七叶皂苷对缺血性脑卒中大鼠血清MBP含量和髓鞘再生的影响 被引量:9

摘要 目的:探讨注射用七叶皂苷对缺血性脑卒中大鼠受损脑髓鞘的保护作用。方法:104只成年雄性SD大鼠,随机分为正常对照组(N)、假手术组(C_-)、大脑中动脉阻塞模型组(C_+)、生理盐水治疗组(E_0)、七叶皂苷(1mg·kg^(-1),qd)治疗组(E_1)。用改良的线栓法制备右侧大脑中动脉阻塞(MCAO)的局灶性脑缺血再灌注模型,用Pal-Weigert髓鞘特殊染色法和ELISA法动态观察各组在1,3,5,7 d时脑髓鞘的再生情况和血清髓鞘碱性蛋白(MBP)的含量。结果:在各个时间点,C_+组内囊有明显的髓鞘脱失,且血清MBP含量也明显升高(P<0.05,P<0.01);E_1组在各个时间点的髓鞘脱失程度明显减轻,且血清MBP含量升高的幅度明显低于C_+组(P<0.01)。结论:注射用七叶皂苷可能通过较明显激发脑的可塑性潜能及其他机制,促进MCAO大鼠缺血损伤区的髓鞘再生。 AIM: To study the protective effects of of focal cerebral ischemia-reperfusion injury. METHODS: aescine for injection on injured cerebral myelin in rats One hundred and four adult male Sprague-Dawley rats were employed in the study, which were randomly divided into normal (N), sham-operation (C-), MCAO model (C+), treated with normal saline (E0), and treated with aescine (1 mg·kg^-1, qd) (E1) groups. Focal cerebral ischemia-reperfusion model was established by middle cerebral artery occlusion (MCAO) on the right side with modified thread embolism method. Each group was continuously observed with remyelination of brain and content of serum myelin basic protein (MBP) on 1 d, 3 d, 5 d, 7 d by Pal-Weigert' s myelin staining and ELISA methods respectively. RESULTS: At appointed time, massive demyelination happened in the internal capsule of C+ group and the content of serum MBP apparently increased (P 〈 0.05, P 〈 0.01 ) . As for El group, the extent of demyelination was lightened distinctly and the content of serum MBP was much lower than that of C+ group (P 〈0.01) at appointed time. CONCLUSION: Aescine for injection could contribute to remyelination of ischemic region of MCAO rats, which maybe related to activate plastic potential and other mechanism in the braiin.
