摘要
应用选择性系列抽提方法与整装细胞制备技术、DGD包埋—去包埋剂切片技术相结合,清晰的显示了BHK_(21)细胞中间纤维,其单丝直径为10nm。近核区的中间纤维呈致密交织的网络,胞质远侧区的中间纤维有规则的平行排列。免疫荧光与免疫胶体金标记实验说明,经上述选择性系列抽提后存留的胞质结构仅对波形蛋白(Vimentin)抗体呈阳性反应。最后用双向电泳技术分析了BHK_(21)细胞中间纤维波形蛋白的分子量为55KD。
By means of serial selective cell extraction, togather with whole-mount cell and DGD embedment-free
section TEM technology, the IFs (intermediate filaments) in BHK cells imaged clearly under electron microscope. The IF unit in BHK cells is 10nm in diametre and its pattern in the cytoplasma distributes polarily. The IFs in the vicinity of the nuclear appear to be a intimate interconnecting network, while which near the border of cell parrel. The results of immunofluorence and immuno-gold electron microscopy in-dicted that the cytoplasmic structure after extraction is only sensitive to antivimentin protein. 2-D gel electrophoresis analysis also suggested that BHK cells are rich in vimentin. All of the results proved that the component of IF system in BHK cells is vimentin, a 55KD protein.
