急性肝损伤时F蛋白含量及HGF和挽肝二号的护肝作用

SERUM F-PROTEIN CONTENT DURING ACUTE HEPATIC INJURY AND PROTECTIVE EFECTS OF HGF AND HEPARTIC DISEASE ⅡON LIVER

应用Ｄ－氨基半乳糖（Ｄ－ＧＬＡＮ）诱发大白鼠急性肝损伤模型，检测了实验动物的血清Ｆ蛋白含量（Ｆ－Ｐｒｏｔｅｉｎ，ＦＰ）、血清总胆红素（ＳＢ）、凝血酶原时间（ＰＴ）、转氨酶（ＡＬＴ）的水平，观察了急性肝损伤的表现并与促肝细胞生长素（ＨｅｐａｔｏｃｙｔｅＧｒｏｗｔｈ－ｐｒｏｍｏｒｉｎｇＦｏｃｔｏｒ，ＨＧＦ）和中药挽肝二号方保护组检测结果相比较。４５只大鼠等分为Ａ、Ｂ、Ｃ３个组。Ａ组由１５只急性肝损伤模型鼠组成，Ｂ组注射Ｄ－氨基半乳糖前先向腹腔注射２ｍｌＨＧＦ，Ｃ组注射Ｄ－氨基半乳糖前３天开始灌胃挽肝２号液。结果：实验肝损伤动物Ａ组即非保护组血清Ｆ蛋白含量和其他三项指标均明显高于Ｂ和Ｃ组（即保护组），在用药２４小时开始变化、４８小时达高峰、７２小时后开始下降；肝病理特点为亚大块坏死，而ＨＧＦ和中药挽肝二号保护组的大鼠血清Ｆ蛋白含量和其他三项指标均显著低于非保护组，肝组织仅见少数肝细胞坏死。因此，可以初步得出结论：Ｆ蛋白是急性肝损伤十分敏感的指标并与急性肝损伤呈正相关，而ＨＧＦ及中药挽肝二号对保护肝细胞，缓解肝坏死有一定作用。

Serum F-protein(FP),total bilirubin(SB),prothrombin time(PT)andtransaminase(ALT)were determined,and acute hepatic injury features were obServedin 45 rats,which were equally divided into 3 groups. the A,B and C group.The Agroup,liver-unprotecting group,consisted of 15 acute hepatic injury model rats,whichwere prepared by injecting Daminogalactose into abdominal cavity in a dose of 900mg/kg. In the B group 2mL hepatocyte growth-promoting factor(HGF)was injected intoabdominal cavity of 15 rats before D-aminogalactose was administered.In the C groupof 15 rats, intragastrical administration of Chinese herbal medicine hepatic diSease Ⅱbegan at the 3 rd day betor injecting D-aminogalactose. The B and C group were calledthe liver-propecting group.The results showed that serum FP,SB,PT and ALT levels inthe B and C group were significantly lower than those in the A group;the hepaticpatholigical features were spotty,picemeal or submassive necrosis in the A group,Whilein the B and C group acute inflammation or a little divergent spotty necrosis was itsnotable characteriStic,It is concluded that F-protein is a quite senitive and specialmarker protein in reflection of the nature and extent of hepatocellular injury,a positivecorrelation existing between FP and hepatocellular in jury;and that HGF and hepaticdisease Ⅱ play a difinite role in propecting hepatocyes and relieving hepatocellular necrosis.
