α-氯丙酸脱卤酶发酵培养基的响应面法优化 被引量:7

Optimization of fermentation conditions for α-chloropropionic acid dehalogenases production based on response surface analysis

摘要 通过Plackett-Burman设计和响应面分析方法对PseudomonasW20菌株产脱卤酶的培养基组成进行优化。得出影响产酶的3个重要的培养基成分为:尿素、葡萄糖和KH2PO4,且其最适浓度分别为1.19、18.4 g.L-1和1.30 g.L-1。此时脱卤酶活力达到了10.57 U.(g干菌体)-1,比优化前提高了23.77%。 Two level factorials designs of Plackett-Burman were constructed to select the Pseudomonas W20 fermentation medium components of a-chloropropionic acid dehalogenases. Three important components were urea, glucose and KHzPO4. The optimized levels of these factors were determined by the response K H2 PO4 23.77% surface analysis. The results showed that the optimum concentrations of urea, glucose and were 1.19 g· L^-1 , 18.4 g · L^-1 and 1.30 g·L^-1 respectively. The enzyme activity increased by under the optimized condition.
