抗原修复对乳腺癌组织中雌激素受体与孕激素受体和C-erbB-2蛋白表达的影响

摘要 目的:探讨使乳腺癌组织雌激素受体(ER)、孕激素受体(PR)和C-erbB-2蛋白在免疫组化中获得最佳效果的抗原修复条件。方法:对100例乳腺浸润性导管癌石蜡标本分别采用高压抗原修复和微波抗原修复法在不同的修复液、不同的时间检测ER、PR和C-erbB-2蛋白的表达,比较和观察它们各自的最优修复条件。结果:高压修复时,ER和PR最适的条件是采用pH值8.0的EDTA修复液煮沸喷气后持续5min,其染色的背景清晰,无非特异性着色。微波修复ER和PR最适的条件是采用pH值8.0的EDTA修复液15min,其染色的背景不清,有非特异性着色。而在高压和微波修复时C-erbB-2蛋白阳性表达率和表达强度的时间最适的条件是分别采用pH值6.0的枸橼酸盐修复液(CA)5min和10min。结论:高压抗原修复法在乳腺癌组织ER、PR和C-erbB-2蛋白阳性表达率、表达强度、背景和非特异性着色等方面均优于微波修复。高压抗原修复法是一种理想有效的抗原修复法,值得推广应用。 Objective To find the best antigen retrieval buffer and retrieval time for the immunohistochemical staining of ER, PR and C-erbB-2. Methods ER, PR and C-erbB-2 protein were detected in different antigen retrieval methods, antigen retrieval buffers, and antigen retrieval time in 100 one hundred breast carcinomas. The positive signals in different conditions were observed and compared under light microscope. Results Using pressure cooker, the best signal for ER and PR was observed after retrieval in the EDTA buffer( pH8.0) for 5 rain. The best signal for ER and PR was observed after retrieval in the EDTA buffer( pH 8.0) for 5 min. The best signal for ER and PR was observed after retrieval in the EDTA buffer( p HS. 0) for 15 min by microwave. The best positive signals for C-erbB-2 were observed after retrieval in the CA buffer( pH 6.0) for 5 min and 10 min by pressure cooker and microwave respectively. Conclusion The high pressure method useful rapid and simple. It is better than that of microwave in many aspects.
