亚洲百合遗传转化受体系统的建立 被引量:11

Establishment of the acceptor system for the gene transformation of Lilium Asiatic hybrids

摘要 以亚洲百合“精粹”(LiliumAsiatic‘Elite’)的鳞片、再生形成的叶片和鳞片叶为外植体,进行了不定芽的诱导分化、试管苗小鳞茎诱导、卡那霉素抗性筛选等研究,建立了亚洲百合遗传转化受体系统。结果表明:鳞片诱导最佳分化培养基为MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,试管苗小鳞茎诱导培养基为MS+IBA 0.5mg/L+70 g/L蔗糖,试管苗叶片分化培养基为MS+TDZ 1.0 mg/L+NAA 0.2 mg/L,试管苗鳞片叶分化培养基为MS+6-BA 2.0 mg/L+2,4-D 0.1 mg/L。抗生素敏感性试验表明,百合叶片的卡那霉素选择压为80 mg/L,鳞片叶为115 mg/L。 Using scales and petioles of Lilium Asiatic hybrid as the study subject, the acceptor system for the gene transformation of Lilium Asiatic hybrids was established. The results showed that the medium suitable for inducing the scales to differentiate Adventitious buds was MS + 6 - BA 2.0 mg/L + NAA 0.2 rag/L, the medium suitable for small bulb induction was MS + IBA 0.5 mg/L + Sucrose 70 g/L, the medium suitable for the leaves of the test-tube plantlets to differentiate adventitious buds was MS + TDZ 1.0 mg/L + NAA 0 2 mg/L, and the medium suitable for the scale leaves of the test-tube plantlets to differentiate adventitious buds was MS+ 6 - BA 2.0 mg/L+ 2,4 - D 0.1 mg/L. The concentrations of the antibiotics for different parts of the test-tube plantlets in their cultures were determined, the concentration of Kanamycin for the petioles was 80 mg/L and for the scales 115 mg/L.
