不结球白菜Pol胞质雄性不育系和其保持系的RAPD分析及分子标记的筛选 被引量:8

摘要 对不结球白菜Pol胞质雄性不育系(CMS)及其保持系基因组DNA进行RAPD分析,共使用了386条随机引物,其中有346条引物在两系之间都得到了扩增产物,64条引物扩增结果在两系之间表现出了遗传多态性。在其不育系中获得了1条稳定扩增的片段OPAY1000,序列测定结果表明,OPAY序列全长为1 053 bp,在GenBank中的登录号为DQ320101。根据序列测定结果设计了1对特异引物,将OPAY1000转化为更稳定的SCAR标记。 The genomic DNA of Pol cytoplasmic male sterility (CMS) line and its maintainer line of non-heading Chinese cabbage was studied by RAPD analysis. Both of lines had obtained amplified products through 346 out of total 386 random primers. They emerged polymorphism in two lines through 64 primers. Furthermore, a specific amplified fragment OPAY1000 of CMS line was found out. As indicated by the sequencing result, OPAY had a 1 053 bp full length, with an accession number DQ320101 in GenBank. According to the sequencing result, one pair of primers was designed to convert OPAY1000 from the RAPD marker into the more stable SCAR marker.
