摘要
利用重组毕赤酵母菌株Swollenin的表达产物——膨胀素,联合果胶酶和纤维素酶组成的复合酶处理茶花粉,经镜检并计算其破壁率.结果表明,单独采用复合酶或重组菌株发酵液对花粉进行破壁处理,破壁率分别为85.7%和89.2%,两者联合处理使茶花粉外壁变软凹陷,萌发沟破裂,内容物呈团状释放出来,花粉的破壁效果明显,破壁率高达92.5%.
The product of the recombinant Pichia pastoris Swollenin strain, which was designated as expansin, or the mixed enzymes of pectinase and cellulase was used to treat the camellia pollen. Result shows that the wall-broken rate of the camellia pollen is 85.7% and 89. 2% as treated respectively with mixed enzymes or the product of recombinant Pichia pastoris Swollenin strain. When treated by mixed enzymes and the product of recombinant strain at the same time. The outer wall of camellia pollen became soft and hollow, the colpus was broken, the matter inside was released, and the wall-broken rate reached 92. 5%. The expansin, a genetic engineered product, and mixed enzymes were applied in wall breaking. The method is an economical and high-efficiency way for the wall-broking of pollen.
