摘要
目的从3株产普鲁兰糖出芽短梗霉As3.3984,As3.837,As3.933中筛选1株产量及糖转化率高、分泌色素低的菌株作为出发菌株。方法相同条件下摇瓶分别培养菌株As3.3984,As3.837,As3.933,测定发酵液黏度并纯化制备普鲁兰糖,比较糖转化率及普鲁兰糖产量;654nm处测定制备的普鲁兰糖溶液的吸光度值,比较色素的分泌量。结果菌株As3.3984普鲁兰糖产量、糖转化率最高,色素含量最低。结论确定菌株As3.3984作为普鲁兰糖生产研究的出发菌株。
Objective To screen a strain with high production of pullulan, high utilization of sugars and low secretion of pigment from As3.3984, As3.837, As3.933. Methods The viscosity of fermentation broths of As3. 3984. As3.837. As3.933 under the same conditions was determined. Pullulan was obtained and the utilized rate of sugars and production of pullulan were observed. A value of the broth of pullulan which was obtained by ethanol deposit was determined at 654 nm for the observation of pigment secretion. Results As3.3984 was the best strain with the highest production of pullulan, utilized rate of the sugars and lowest secretion of pigment. Conclusion As3.3984 can be used as the original strain for the production of pullulan.
