一氧化氮在兔生长板软骨细胞终末分化中的生物学作用 被引量:2

摘要 原代分离及培养兔肋软骨生长板软骨细胞,添加不同浓度的全反式维甲酸(AT-RA)诱导软骨细胞的终末分化。酶动力学法检测碱性磷酸酶(AKP)及细胞培养上清液中一氧化氮合酶(NOS)活性;硝酸还原酶法检测所培养软骨细胞上清液中一氧化氮(NO)浓度;AT-RA分别联合NOS抑制剂L-硝基精氨酸甲酯(L-NAME)及可以产生NO的S-亚硝基谷胱甘肽(SNOG)作用于原代培养的生长板软骨细胞,分别检测AKP活性。结果发现随着AT-RA浓度的递增,AKP、NOS活性及NO浓度明显增加(P<0.05);L-NAME明显抑制所培养的生长板软骨细胞AKP的活性(P<0.05),SNOG可以提高所培养的生长板软骨细胞AKP的活性(P<0.05)。认为NO可以促进兔肋软骨生长板软骨细胞的终末分化。
