摘要
目的观察不同剂量地塞米松对SIRS大鼠心肌酶CK-MB的影响。方法Wistar大鼠40只,体重160~200g。雄性。随机分为5组:LPS注射后3h组(3.0h组)、LPS注射后6h组(6.0h组)、小剂量地塞米松治疗组(DXMI组)、大剂量地塞米松治疗组(DXMⅡ组)、正常对照组,每组8只。实验组大鼠给予大肠杆菌内毒素(E.Coli O55B5,美国Sigma公司产品)20mg/kg腹腔注射,制造SIRS大鼠模型,观察时间分别为3、6h,DXM组在同样给予LPS后10min给地塞米松,Ⅰ组给予10mg/kg,Ⅱ组给予20mg/kg腹腔注射,观察时间为3h,结果与LPS3.0h组进行对比,对照组腹腔注射等量无菌生理盐水。观察SIRS大鼠各个时间心肌酶谱CK-MB的变化。结果LPS3.0h组、LPS6.0h组分别与对照组比较,结果有显著性差异(P均〈0.01),尤以LPS6.0h组升高明显,说明SIRS大鼠心肌细胞受到严重损害。而且随着时间的推移,损害越来越重。与LPS3.0h组比较,DXMI组CK-MB值下降明显(P〈0.05),DXMⅡ组CK-MB值略有下降,但无显著性差异(P〉0.05)。结论SIRS大鼠心肌受到损害,大剂量地塞米松对SIRS大鼠心肌损害的改善不明显,小剂量地塞米松对SIRS大鼠心肌损害的疗效优于大剂量地塞米松。
Objective To explore the effects of different doses of dexamethason on systemic inflammatory response syndrome rat's cardiac CK-MB. Methods Forty Wistar rats were randomly divided into five groups: LPS 3.0 h grouD LPS 6.0 h group, DXM Ⅰ group, DXM Ⅱ group, control group. The SIRS model of rats was made by lipopolysaccharide (LPS) injection, and DXM Ⅰ group received DXM 10 mg/kg, DXM Ⅱ] group received DXM 20 mg/kg, intraperitoneally. The cardiac CK-MB of each group were observed and compared. Results Compared with the control group, the CK-MB of LPS 3.0 h group, LPS 6.0 h group were significantly increased ( all P 〈0.01 ), Compared with LPS3.0 h group, the CK-MB of DXM I group significantly decreased(P〈 0.05), but DXM I] group did not obviously decreased (P〈0.05). Conclusion The effects of 10 mg/kg dose of dexamethason was better than 20 mg/kg dose of dexamethason on improvement of SIRS rat' s cardiac CK-MB.
