摘要
目的:观察珍珠丸对兔视网膜缺血再灌注损伤Bcl-2基因表达的影响,探讨珍珠丸对视神经的保护作用。方法:采用升高兔眼压到14.63Kpa持续1小时的方法制作视网膜缺血再灌注模型。将35只日本大耳白兔随机分为正常组、缺血再灌注组及治疗组。采用光镜观察再灌注后不同时间段视网膜组织学变化,运用免疫组化SABC法观测不同时间段治疗组与对照组视网膜组织中Bcl-2基因的表达,进行图像分析。结果:光镜下,缺血再灌注组神经节细胞比正常组明显减少,空泡形成和核固缩现象多见,内网状层、内核层轻度变薄,内核层细胞排列紊乱,外核层变化不明显。Bcl-2基因表达有所增高。而珍珠丸组内核层、神经节细胞层厚度及神经节细胞数目与正常组比较无明显差异,内核层和节细胞层细胞排列比较规整。Bcl-2基因表达量比缺血再灌注组明显增加(P<0.05),与维生素E组比较有显著性差异(P<0.05),但168小时后缺血再灌注组与正常组比较无显著性差异,各组两个时间段Bcl-2基因的表达均有显著性差异。结论:珍珠丸能减少神经元细胞凋亡的数目,并能促成Bcl-2基因的表达,有保护神经元细胞的作用。对于视网膜缺血再灌注损伤具有保护作用,提示珍珠丸可能用于临床上视网膜缺血再灌注损伤的治疗。
Objective: To investigate the zhenzhuwan' s neuroprotection to optic nerve through observe the influence of the zhenzhuwan to express of Bel- 2 in rabbits' isehemal reperfusion.Methods: Raising zop to 14.63kpa for one hour was carried out tomake the mold of retinal ischemal , 35 Japan big - ear rabbits were divided into normal group , ischemal group and treated group by chance. Observe the change of retinal histology in different periods after reperfusion by optics microscope. Study the expess of Bcl - 2 in different periods through immunohistochemistry SABC method. Analysis the icon. Results: In optic microscope, the ganglion cell of ischemal reperfusion group were obviously less than normal group, cavitation and pycnosis were more than it, plexiform layer and inner nuclear layer changed thinner, inner nuclear layer cell arranged disorder, outer nuclear layer had no significant difference. Bcl - 2 become more obvious. But in treated group, inner nuclear layer the thickness of ganglion - cell layer and the number of ganglion cells had no significant difference to normal ones, the inner nuclear layer and ganghon - cell layer arranged neat. Bcl - 2 had significant difference. Conclusion: Zhenzhuwan can reduce the number of Apoptosis of neuron - cyte, can protect neuron - cyte; Zhenzhuwan may be used to protect the damage of icon reperfusion of retine in clinic.
