Determination of Ketorolac in Human Urine by HPLC

摘要 目的建立以高效液相色谱法测定人尿中酮洛酸浓度的方法。方法色谱柱为Waters Sunfire C18,流动相为0.02mol·L-1磷酸二氢钾溶液-乙腈(70∶30,用磷酸调节pH值至4.5),检测波长为315nm,柱温为35℃,流速为1mL.min-1,进样量为20μL,保留时间为9.1min。结果人尿中酮洛酸浓度在0.1~10.0μg·mL-1范围内线性关系良好,日内和日间RSD<10%,最低定量限为0.1μg·mL-1。结论本法简便、快捷、灵敏,适用于酮洛酸人体药动学研究。 OBJECTIVE:To establish a HPLC method for the determination of ketorolac in human urine.METHODS:The chromatographic column was Waters Sunfire C18, the mobile phase consisted of 0.02mol · L^-1 potassium dihydrogen phosphate- methyl cyanide(70 : 30, adjusted to pH4.5 with phosphoric acid) .The detection wavelength was 315nm; column temperature was 35℃ ; the flow rate was 1mL · min^-1 The sample size was 20μL and retention time was 9. 1min. RESULTS: The linear range of ketorolac concentration in human urine was 0.1- 10.0μg · mL^ -1,The inter - day RSD and intra - day RSD were less than 10% .The minimum quantity limitation was 0. 1μg · mL^-1.CONCLUSION:This method is a sensitive, convenient and feasible for ketorolac detection.
