摘要
目的:探讨5-氟尿嘧啶(5-Fu)对人结肠癌细胞凋亡及其相关基因表达的影响。方法:Hct/8细胞分为5-Fu组和对照组,通过凋亡细胞数量、形态、基因组DNA片段化及流式细胞仪检测观察5-Fu对人结肠癌细胞凋亡情况的影响,并通过基因芯片测定相关基因改变。结果:吖啶橙染色及基因组DNA电泳显示5-Fu组凋亡细胞数量明显多于对照组(P<0.01);基因芯片检测显示,5-Fu致多种基因表达发生改变,尤其是与DNA损伤修复相关基因出现表达下调。结论:5-Fu具有致人结肠癌细胞凋亡的作用,其原因是由于多种基因表达改变相互作用影响所致,尤其是DNA损伤修复相关基因的改变致不能及时修复受损的基因,最终致癌细胞发生凋亡。
Objective To explore the effects of 5-Fu on apoptosis of colon cancer cells (Hct/ expressions. Methods Hct/8 were divided into 5-Fu group and control group. The apoptosis 5-Fuwas observed using cell counting, morphology , DNA segmentation and flow cytometry, gene expression was analyzed by microarray chip. Results The number of apoptotic ceils was 8) and related gene of Hct/8 treated by and the changes of greatly increased in 5 Fu group compared with control group (P〈 0.01), and further comfirmed by aradine staining and genomic electrophoresis. Microarray chip analysis showed that treatment of 5-Fu on Hct/8 cells resulted in changes of various gene expressions, especially dowwregulated of DNA injury-repair related genes. Conclusion 5-Fu could induce the apoptosis of Hct/8 due to interaction of various gene expression changes, particularly DNA injury-repair related genes could not repair the damaged gene in time.
