天麻素对体外模拟脑缺血损伤大鼠脑微血管内皮细胞的保护作用 被引量:29

摘要 目的:观察中药天麻素对培养的正常及缺血大鼠脑微血管内皮细胞(BMEC)的影响,以探讨天麻素对BMEC的作用。方法:采用体外培养的BMEC,模拟脑缺血损伤,观察天麻素对其活性、生存率和一氧化氮(NO)的影响。结果:缺血损伤模型BMEC活性及生存率较正常对照组明显降低;不同剂量天麻素对正常对照组BMEC活性、生存率及NO含量有升高趋势;正常对照组不同剂量天麻素BMEC活性较缺血损伤模型组活性有明显升高(P<0.05);天麻素高剂量可明显提高缺血损伤BMEC NO含量(P<0.05)。结论:天麻素各实验剂量可增强缺血损伤BMEC活力,提高受损内皮细胞(EC)的生存率;促使EC NO分泌增加。提示天麻素对体外培养的大鼠BMEC缺血损伤具有一定的保护作用。 Objective: To observe the protective effects of Gastredine on the cultivated rat brain mierovessel endothelial cells damage by mimic cerebral ischemia. Methods: The endothelial cells activity, survival rate, the change in NO content, and the effects of Gastrodine were observed in the cultivated rat brain microvessel endothelial cells (BMEC) damaged by mimic cerebral ischemia. Results: The activity and survival rate of BMEC in the ischemia goups are obviously lower than that in the normal goups; compared with the normal groups, the activity, survival rate and NO content of BMEC in the Gastredin groups have the increasing tendency; comparing to the ischemia groups, the activity of BMEC in the Gastredin groups obviously increasing ( P 〈0.05); NO content of ischemia injured BMEC in the high dose Gustredin goups increases obviously ( P 〈0.05). Conclusion: The Gastredin at suitable does could enhance the activity of ischemia injured BMEC and increase survival rate of injured EC and NO secretary volume of EC. It suggests that the gastredin plays the protection role in ischemia injured rat BMEC cultured in vitro.
