摘要
目的 通过尾加压素Ⅱ受体桔抗剂(URA)对急性肺损伤(ALI)大鼠的干预,检测其支气管肺泡灌洗液中尾加压素Ⅱ(UⅡ)的浓度,探讨URA对早期ALI的影响。方法 42只SPF级Sprague Dawley(SD)大鼠随机分为2组,各21只。所有大鼠均用油酸复制ALI动物模型。随机选取其中一组为ALI模型组,另一组加注射URA为干预组。每组分别于3、12、24h后活杀并取右肺行支气管肺泡灌洗,留取支气管肺泡灌洗液(BALF)。BALF离心后留取上清液置于-80℃冰箱待作UⅡ测定。结果 ALI模型组3、12、24hBALF中UⅡ值分别为(73.8±7.0)、(30.7±5.3)和(14.7±1.9)ng/L;干预组分别为(21.5±3.5)、(63.1±6.5)和(79.0±8.8)ng/L,随着肺损伤时间的延长,ALI模型组大鼠BALF中UⅡ进行性降低(P〈0.05),URA对干预组BALF中UⅡ水平有明显影响(P〈0.05)。结论 URA可能通过抑制UⅡ的作用而对早期ALI产生保护作用。
Objective To investigate the alteration of urotensin Ⅱ of bronchoalveolar lavage fluid (BALF) in rats of acute lung injury(ALI), affected by the urotensin Ⅱ receptor antagonist(URA) ,thereby to understand the function of URA to ALL Methods Forty-two Sprague Dawley ( SD ) rats were divided into two groups randomly ( n = 21 for each group). All rats were injected into oleic acid to establish ALI models. G1 was taken as ALI model group and the other group were injected with URA as experiment group (G2). The rats were sacrificed and BALF was drawn in 3,12 ,and 24 hours. The upper fluid was separated from the BALF and was kept in -80 ℃ for being detected. Resuits The concentration of U U of G1 in 3,12,24hours was ( 73.8 ± 7.0) pg/ml, ( 30.7 ± 5.3 ) pg/ml, and ( 14.7 ± 1.9 ) pg/ml,respectively ; and ( 21.5 ± 3.5 ) pg/ml, ( 63.1 ± 6.5 ) pg/ml, and ( 79.0 ± 8.8 ) pg/ml in the same time of G2. With the time, the UⅡ concentration in BALF was significantly decreased ( P 〈 0.05 ) in G1 and increased significantly ( P 〈 0.05 ) in G2. Conclusion URA may protect the ALI at the early stage by inhibiting the effect of U Ⅱ.
