摘要 RNA干扰(RNAi)技术是一种发展迅速并极富应用前景的基因控制技术,通过产生内源性特异序列的小干扰RNA(siRNA),诱导特异靶基因mRNA降解,从而使该基因“沉默”。转化生长因子(TGF)-β1是器官纤维化过程中的中心介质,能促进纤维母细胞的大量复制,刺激细胞分化或基质合成,增加细胞外基质的聚积并抑制其降解,在细胞间质产生胶原沉积。应用RNAi技术抑制相关基因的表达来防治纤维化疾病已成为可能。 RNA interference (RNAi) is a genie technology which has been named "genie silencing", the mechanism is that small interference RNA triggers the degradation of homologous mRNAs. RNAi has been shown to represent a powerful method for manipulating gene expression in orgamisms and is developed into a promising therapeutic tool. Transforming growth factor-beta-1 ( TGF-β1 ) is a central transmitter in the process of organic fibrosis. TGF-β1 can enhance fibroblasts' multiplication, stimulate cytodifferentiation or metrical compostion, enhance aggregation of the rnetrix and inhibit its degradation, lead to deposition of collaten in the intercellular substance. It is possible that target genie expression has been inhibited by RNAi.
