重组人干扰素γ对球后成纤维细胞CD40 mRNA表达的影响 被引量:2

The Effects of rhIFN-γ Induced CD40 mRNA Expression in Cultured Human Retro-ocular Fibroblasts

摘要 目的探讨在重组人干扰素γ(rhIFN-γ)作用下,正常人和甲状腺相关眼病(TAO)患者球后成纤维细胞(RF)CD40 mRNA的表达情况。方法用RT-PCR法,对不同浓度rhIFN-γ及400 U/mL rhIFN-γ作用不同时间诱导体外培养的正常人和TAO患者RF CD40 mRNA进行半定量分析。结果经100,200,400,1 000 U/mLrhIFN-γ作用48 h后,正常人和TAO患者RF CD40的光密度比值与各自对照组比较,差别有统计学意义(P<0.01);两者同一浓度比较差别无统计学意义(P>0.05)。经400 U/mL rhIFN-γ作用12,24,48,72 h后,正常人和TAO患者RF CD40的光密度比值与各自对照组比较差别有统计学意义(P<0.05),两者同一时间比较差别无统计学意义(P>0.05)。结论正常人和TAO患者RF均表达CD40 mRNA。rhIFN-γ可上调正常人和TAO患者RF CD40 mRNA的水平,呈浓度和时间依赖。 Objective To quantify the mRNA expression of CD40 induced by recombinant human interferon gamma(rhIFN-γ) in cultured human retro-ocular fibroblast(RF) from normal subjects and thyriod associated ophthalmophy (TAO) patients. Methods Half quantifying the mRNA expression of CD40 induced by different concentration and time rhIFN-7 in cultured human RF from normal subjects and TAO patients. Results The absorbency values of 100, 200, 400, 1 000 U/mL rhIFN-γ in RF from normal subjects and TAO patients were increased significantly than negative control and 50 U/mL rhIFN-γ group(P〈0.01). Both not difference in same concentration. The absorbency values of 12,24,48,72 h of 400 U/mL rhIFN-γ in RF from normal subjects and TAO patients, showed significantly different compared with negative control group(P〈0.01) ,and no difference in same time. Conclusions The cultured human RF from normal subjects and patients with TAO all express CD40 mRNA. The rhIFN-γ could up-regulate CD40 gene mRNA expression with dose-dependence and time-dependent in both human RF.
