白花蛇舌草注射液诱导人肺癌细胞株SPC-A-1凋亡及其分子机制的研究 被引量:29

摘要 目的探讨白花蛇舌草注射液诱导人肺癌SPC-A-1细胞凋亡及其分子机制。方法采用MTT法检测癌细胞增殖抑制率,流式细胞仪检测细胞凋亡率,免疫组化检测野生型P53、Bcl-2和NF-κB(P65)蛋白表达。结果白花蛇舌草注射液能抑制SPC-A-1细胞增殖,且呈剂量依赖性;DNA直方图上呈现凋亡细胞所特有的亚二倍体峰(sub-G1),5和50ml/L白花蛇舌草注射液组细胞凋亡显著升高,P53蛋白表达显著上升,Bcl-2和NF-κB蛋白表达显著下降。结论白花蛇舌草注射液能诱导人肺癌SPC-A-1细胞凋亡,其机制可能与上调P53蛋白表达以及下调Bcl-2和NF-κB蛋白表达有关。 Objective To explore the effect of Hedyofic diffnsa Willd injection (HDI) on apoptosis of human lung cancer SFC-A-1 cells and its molecular mechanism. Methods The growth inhibitory rate was evaluated by MTT assay. Apoptotic rate was determined by flow cytometry. The expression of wild type P53, Bcl-2 and NF-κB ( P65 ) proteins were detected by immunohistochemical staining, Results HDI could inhibit the proliferation of SPC-A-1 ceils and inhibitory effect was dose-dependent. SPC-A-1 cells shoved the peak of sub-G1 on DNA histogram. The apoptotic rate of 5 and 50 ml/L HDI groups were increased markedly. The expression of P53 protein was increased obviously while Bcl-2 and NF-κB ( P65 ) proteins were decreased significantly. Conclusions HDI can induce the apeptosis of SPC-A-1 cells, and its molecular mechanism may be associated with the up-regulating expression of P53 protein and down-regulating expression of Bc1-2 and NF-κB proteins.
