摘要
采用不同培养基配方对5个不同来源的白灵菇菌株,在液体和固体培养条件下研究了其菌丝生长的生物学特性。结果表明,不同氮源对菌丝生长有明显的影响,以豆粉、蛋白胨作氮源最佳,其次是麦麸即碳源,玉米粉和牛肉膏不适宜作为白灵菇菌丝生长的氮源。5个菌株在PDA、无机盐组合培养基上菌丝的生长速率明显不同。在PDA上,菌丝生长的快慢依次为天山2号、L1、白灵菇1号、W1W2和江都菌株。不同配方对菌丝生长的影响不同,固体培养条件下,B4、B3、B2、B1、B11配方比较适合于各菌株生长,但来自于江都的白灵菇菌株表现出对无机盐类有较强的适应性,菌丝生长较其它菌株快,日均生长率达10 mm以上;液体培养条件下,B9、B14和B16配方的菌丝生物量最大,其次是B1、B3、B5配方。表明,固体培养下的配方不一定完全适合于作液体培养基配方。5个菌株的对峙实验表明,白灵菇1号与L1菌株间亲缘很近,江都菌株与白灵菇1号、天山2号菌株间亲缘关系较远,而与L1、W1W2菌株间亲缘关系比较近。W1W2菌株与白灵菇1号、天山2号、L1、江都菌株间的亲缘关系较近。
Five different Pleutotus nerbrodensis strains were cultured on the liquid or solid medium with different medium formula to investigate effects of different medium formula on hyphal growth of Pleutotus nerbrodensis. The results show that the different nitrogen sources significantly affect the hyphal growth, the soybean flour and peptone are the best and the wheat bran is better for the hyphal growth, and the maize meal and beef extract are not suitable for the hyphal growth. The hyphal growth rate of five strains on PDA or inorganic salt combination media is the obvious difference and the hyphal growth rate of five strains cultured on PDA is Tianshan 2〉 L1〉 Bailing 1〉W1W2〉Jangdu strain. The different medium formula has the different effects on the hyphal growth. The B4, B3, B2, B1 and B11 formulas are suitable for all tested strain's hyphal growth under solid media condition. The Jangdu strain is of the strong growth adaptability on the inorganic salts and its hyphal growth rate with above 10 mm/day is more quick compared with other strains. The hyphal biomass of B9, B14 and B16 formulas is up to maximum under the liquid culture condition, which indicates that the formulas of solid culture does not completely fit in liquid formulas. The results show that the relationship between Bailing 1 and L1 strain is very close, the relationship between W1W2 and Bailing 1, Tianshan 2, L1, Jiangdu strain is close. The relationship between Jiangdu and Bailing 1, Tianshan 2 strain is comparative far, but the relationship between Jiangdu and L1, W1W2 strain is very close.
