摘要
目的采用HPLC—ELSD法测定不同产地桑叶中1-脱氧野尻霉素的含量。方法色谱柱为TSKgel Amide-80(4.6mm×250mm,5μm);流动相为乙腈-水(70:30),水中含有10mmol/L醋酸铵;流速1.0ml/min。结果线性范围为2.11~10.55μg,r=0.9991。平均回收率为95.98%(n=5),RSD=1.98%。结论本方法快速、简便、重现性好,可用于桑叶及相关样品中DNJ的定量分析。
Objective To determine the content of 1 - deoxynojirimycin in Morus alba L.leaves from different areas using HPLC - ELSD.Methods A HPLC method was established using TSKgel Amide- 80(4.6 × 250 mm,5μm)with acetonitrile- water (70: 30) as mobile phase and the water containing 10 mmol/L ammonium acetate. The flow rate was 1.0 ml/min.Results The calibration curve was linear in the range of 2.11~10. 55μg,r=0.9991. The average recovery was 95.98%( n=5), RSD=1.98%.Conclusions The method is rapid, simple, stable and can be used for the determination of 1 - deoxynojirimycin in mulberry leaves and other related products.
