摘要
目的:探讨胸苷酸合成酶(TS)及二氢嘧啶脱氢酶(DPD)转录水平及其对5-氟脲嘧啶疗效的预测价值。方法:利用逆转录聚合酶链反应(RT-PCR)方法从胃肠癌细胞株中扩增胸苷酸合成酶及二氢嘧啶脱氢酶的DNA片段,并与内参照基因GAPDH进行比较。结果:对5-FU敏感的细胞株MKN45 TS和DPD的表达水平较低,而其他对5-FU不敏感的细胞株则无此现象。结论:TS及DPD mRNA水平对应用5-氟脲嘧啶治疗胃肠癌的疗效具有一定预测价值。
Objective To investigate the effect of expression levels of TS and DPD of tumor cells on the treatment efficacy of 5 - flurouracil. Methods The expressions of TS and DPD mRNA in 6 strains of cancer cells (gastric 3, colorectal 3) were examined with RT- PCR. Expression of GAPDH was examined for contrast. Results It was demonstrated that only with the chemotherapy - sensitive gastric cancer cell strain MKN45, the expressions of TS and DPN were obviously lower than those in other 5 cancer cell strains. Conclusion Examination of TS and DPD mRNA expressions in tumor specimens might provide some prediction of 5Fu treatment efficacy.
