苦参碱抑制HepG2增殖时Ca^(2+)等生化指标的变化 被引量:4

摘要 目的:研究苦参碱抑制HepG2细胞增殖时相关生化指标的变化,有助于认识苦参碱抗肿瘤的机制。方法:一定浓度苦参碱处理HepG2细胞,Fluo-3/AM荧光染色,荧光显微镜观察细胞内Ca2+浓度变化;免疫细胞化学和放免法检测AFP、PCNA、cAMP、cGMP的改变;亲和层析法分离肝癌GGT(HSGGT),酶动力法检测HSGGT和GGT酶活性。结果:苦参碱抑制HepG2细胞增殖时,胞内Ca2+浓度升高;反应细胞生长抑制的指标—cAMP升高,cAMP/cGMP比值升高。反应细胞增殖的生化指标—AFP、PCNA、cGMP、GGT、HSGGT量均降低。结论:苦参碱抑制HepG2细胞增殖时,相关生化指标均发生了改变,这可能与苦参碱抗肿瘤的机制有关。 Objective:To understand the anti-tumor function of Matrine ,we examined the changes of biochemistry factors in HepG2 cells after inhibited by Matrine. Method:Different doses of Matrine treated HepG2, stained the cell with Fluo 3/AM, and checked Ca^2+ under the Laser Scanning Confocal Microscope. Changes of AFP,PCNA,cAMP and cGMP were deteeted with immunoeytoehemistry and radioimmunoassay; hepatoeellulors' GGT (HSGGT) was isolated with affinity chromatography;enzymatie activation of HSGGT and GGT were detected with enzymatic assay. Result:In treated cells, the concentration of Ca^2+ inereased; the index of cells growth inhibition -cAMP rose and the ratio of cAMP to cGMP rose; the index of cells proliferation-AFP,PCNA,eGMP.GGT,HSGGT-all fell. Conclusion :Correlated Biochemistry factors change in HepG2 cells after treated by Matrine,which may be relative to the antitumor funetion of Matrine.
