摘要
目的:探讨鱼藤素对DNA拓扑异构酶TopoⅠ表达的影响,进一步明确鱼藤素抑制肿瘤细胞增殖的作用机制。方法:流式细胞术及RT-PCR法分析鱼藤素作用K562细胞后TopoⅠ蛋白及mRNA含量的变化。结果:流式细胞术分析结果发现TopoⅠ蛋白的平均荧光强度随着鱼藤素作用浓度的增高而逐渐减少,与空白对照组比较有显著差异(P<0.05);RT-PCR半定量检测结果显示,鱼藤素作用K562细胞引起TopoⅠmRNA含量减少,并与其浓度正相关。结论:鱼藤素可通过抑制肿瘤细胞内TopoⅠ而发挥作用。
Objective: To investigate the effect of deguelin on the expression of TopoⅠ in K562 cells. Methods: Changes in TopoⅠ mRNA and protein levels were ohserved by RT-PCR and flow cytometry. Results: Flow cytometry showed that gradually decreasing expression of the TopoⅠ protein was induced by an increase of deguelin protein. RT-PCR analysis also showed that deguelin reduced TopoⅠ mRNA in a dose-dependent manner. Conclusion: Deguelin can inhibit the expression of TopoⅠ protein. The anti-proliferative activity of deguelin is related to TopoⅠ protein expression in K562 cells.
