MAPK反义寡核苷酸对乙醛活化HSC增殖和JNK信号通路的影响

Effects of mitogen-activated protein kinase antisense oligodeoxynucleotide on activation of acetaldehyde-induced hepatic stellate cell proliferation and JNK signal pathway

摘要 目的:探讨丝裂原活化蛋白激酶(MAPK)反义寡核苷酸(ASO)对乙醛活化肝星状细胞(HSC)增殖和MAPK中的c-Jun氨基末端激酶(JNK)信号通路的影响。方法:用脂质体转染法将MAPKASO导入乙醛活化HSC中,通过MTT法检测细胞增殖情况、流式细胞仪检测MAPK-JNK表达。结果:MAPK ASO可明显抑制细胞增殖及抑制JNK信号通路的表达,其抑制率分别为40.81%、60.6%。与对照组比较有显著差异(P<0.05)。结论:MAPKASO能够抑制HSC的增殖,阻断JNK信号通路,这可能成为治疗肝纤维化的一个有效途径。 Objectives:To study the effects of mitogen - activated protein kinase antisense oligodeoxynudeotide (MAPK ASO) on the activation of acetaldehyde- induced hepatic stellate cell (HSC) proliferation and JNK signal pathway. Method: After MAPK ASK) was introduced into activated HSC by means of liposome transfection, the proliferation of HSC was evaluated by MTT assay. The expression of JNK pathway was examined by flow cytometry. Results: Compared with the control group, the HSCs proliferation and JNK signal pathway in the experimental group were significantly inhibited hy MAPK ASO and the inhibitory rate was 40. 81% and 60. 6%, respectivdy (P〈0. 05). Conclusion: MAPK ASO intercepts JNK signal pathway hy inhibiting HSC proliferation, which maybe hecome an effective therapy for liver fibrosis.
