摘要
大孔硅胶与乙烯基硅烷反应后,再与甲基丙烯酸羟乙基酯和二乙烯基苯共聚成一种新型分高蛋白质的反相色谱填料。在这种色谱填料上考察了对蛋白质及实际样品的分离能力,它具有柱效高、惰性好和分离效率高的优点,为生物大分子的高效液相色谱快速分离提供了一种新的手段。
A novel reversed-phase chromatographic packing for protein separation is described. First diethoxymethylvinyl silane was bonded onto silicagel support on which hydroxyethyl methacrylate and divinylbenzene were then copolymerized to produce a packing with network-structured surface. This packing has -high column efficiency, good resoiving power and inertness. A baseline-separation of a model protein mixture of lysozyme. eytochrome C, β-lactoglobulin, chicken egg albumin, ribonuclease and pepsin is achievable within 15 min. It is likely that this macroporous silicagel polymer bonded packing can be used to separate biopolymers quickIy. The application of this packing to the purification of casein demonstrates its capability of chromatographic preparation.
