摘要
采用四氢呋喃∶水∶乙腈∶乙酸(50∶30∶20∶1,V/V)混合液提取马铃薯中总配糖生物碱(TGA)。离心后,上清液在酸性条件下超声波处理,氨水调节pH10.5,沸水浴蒸干,用pH10.5的氨水洗涤残留物,离心。7%磷酸溶解TGA。室温下,加0.03%聚甲醛磷酸溶液显色,30min后,于620nm波长处测定吸光度值,标准曲线法定量。对α-茄减的回收率为100%~105%。
A procedure for deterrnining total gly-coalkaloids of potato is descrided in the pre-sent paper.Smashed potato was extracted withTFA-H_2O-CH_3CN-HAc[50∶30∶20∶1,(V/V)] solution and then centrifuged. The su-pernatant was subjected to ultrasonic treat-ment under acidic condition,and the pH wasadjusted to pH 10.5 with NH_3·H_2O. Waterbath was used to evaporate the solution,thenthe residue was washed twice with NH_3·H_2O(pH10.5)and dissolved in 7% H_3PO_4 solu-tion. Solution of 0. 03%paraformaldehyde wasused for color deveIopment at room tempera-ture for 30min,the absorbance was measuredat 620nm.Theresults indicated that therecov-erv of α-solanine was 100-105%.
