摘要
目的:探讨1型嗜肺军团菌(LP 1)与其他常见细菌性肺炎病原菌之间是否存在共同抗原。方法:用LP 1菌体抗原免疫家兔,取其血清作为多克隆一抗;提取LP 1和肺炎链球菌等细菌的菌体抗原;将这些抗原进行聚丙烯酰胺凝胶电泳(SDS-PAGE);再进行免疫印迹分析(W estern-b lot)。结果:W estern-b lot显示:表皮葡萄球菌、流感嗜血杆菌和铜绿假单胞菌在154 kD a和97 kD a处、流感嗜血杆菌和铜绿假单胞菌在79 kD a处、大肠埃希菌和流感嗜血杆菌及肺炎克雷伯菌在58 kD a处、铜绿假单胞菌在44 kD a处与LP 1多抗均有交叉反应。金黄色葡萄球菌和肺炎链球菌与LP 1多抗未出现结合反应。结论:LP 1和流感嗜血杆菌、铜绿假单胞菌之间存在有3个具有交叉反应的膜抗原条带,和表皮葡萄球菌之间有2个,和大肠杆菌、肺炎克雷伯杆菌之间只有1个,和金黄色葡萄球、肺炎链球菌间没有交叉反应的膜抗原条带。
Objective.To study whether there were the same antigens between Legionella pneumonia serogroup 1 and the other pathogenic bacteria of pneumonia. Methods :Two rabbits were immunized with the somatic antigens of Legionella pneumophila serogroup 1 (LP1),the serum were separated from the whole blood used as the anti- LP1 antibody. The bacterial somatic antigens were extracted including Legionella pneumophila sergroup 1 (LP1) and Bacillus coli,and so on. All these antigens were separated by SDS-PAGE gel electrophoresls ,then tested by Western-blot assay. Results: The Western-blot assay showed that there were the same bands in the lanes of Staphylococcus epidermidis,hemophilies infl,:enzae and Pseudomonas pyocyanea at the position of 154 kDa and 97 kDa ,in the lanes of hemophilies influenzae and Pseudomonas pyocyanea at the position of 79 kDa,in the lanes of Bacillus eoli,hemophilles influenzae,Bacillus eanalls capsulatus at the position of 58 kDa and in the lane of Pseudomonas pyocyanea at the position of 44 kDa:there were no bands in the lanes of Staphylococcus aureus and Streptococcus pneumoniae. Conclusion :There are 3 same antigen bands in the bacterial somatic antigens among LP1,Hemophilies influenzae,Pseudomonas pyocyanea and LPl;There were 2 ones between LP1 and Staphylococcus epidermidis ,and only one among LP1 and Bacillus eoli ,Bacillus canalis capsulatus, and nothing among LP1 and Staphylococcus aureus,Streptococcus pneumoniae.
