前列腺癌中COX-2 TGF-β1表达与肿瘤发生浸润和转移的相关性研究 被引量:1

摘要 目的:通过检测前列腺癌(PCa)、癌前期病变及良性前列腺增生(BPH)组织中COX-2、TGF-β1表达,探讨其在PCa发生、发展及转移中的作用。方法:随机选择39例PCa、13例癌前期病变和23例BPH标本,应用免疫组化SABC法分别检测COX-2和TGF-β1的表达强度、分布和形态,并采用北航病理图像分析系统分析实验结果。结果:在BPH和PCa组织中,COX-2阳性率分别为8.70%(2/23)、64.10%(25/39);而TGF-β1的阳性率分别为21.74%(5/23)、79.49%(31/39)。COX-2、TGF-β1在PCa组织中的表达均明显高于BPH(P<0.01)。结论:COX-2、TGF-β1表达与PCa生物学行为关系密切,尤其在PCa浸润、转移过程中,二者可能通过促进PCa细胞增殖、抑制细胞凋亡和诱导血管生成等协同发挥重要作用。 Objective:To study the expression of COX-2 and TGF-β1 protein and its correlation in neoplasia, invasion and metastasis in prostatic carcinoma(PCa). Methods : 75 cases of prostate specimens including prostatic adenoearcinoma, precancerous lesions and benign prostatic hyperplasia(BPH) were collected randomly, and SABC method was used to evaluate the positivity of COX-2, TGF-β1 protein in the specimens. Results : The positive expression rates of COX- 2 and TGF-β1 protein were 8.70% (2/23)and 21.74% (5/23)in BPH while that was 64. 10 % (25/39)and 79.49% (31/39)in PCa, respectively. The expressions of COX-2 and TGF-β1 in PCa were significantly higher than that in BPH( P 〈 0.01 ). Conclusion : Over expression of COX-2 and TGF-β1 are positively correlated with the clinical stage and grade of PCa. COX-2 may cooperate with TGF-β1, which plays an important role in the carcinogenesis and progression of PCa by promoting cell proliferation,inhibiting cell apoptosis and inducing tumor angiogenesis.
