大肠癌中nm23-H1的表达及其与临床病理特征的关系 被引量:5

Expression of nm23-H1 and its relationship with clinicopathological characteristics in human colorectal carcinoma

摘要 目的:观察nm23-H1的表达与临床病理特征的关系,探讨nm23-H1的表达与大肠癌发生、发展、转移的作用.方法:选取行结、直肠癌根治术的患者63例,并取腺瘤组织16例做对照.应用免疫组化S-P法检测大肠癌患者癌组织、同源癌旁组织及正常黏膜组织和大肠腺瘤组织nm23-H1的表达.结果:nm23-H1在癌组织中表达(58.7%)显著低于癌旁组织(90.5%)、正常黏膜组织(96.0%)及腺瘤组织(93.8%)(P<0.05).在大肠癌组织中,无淋巴结转移者nm23-H1表达(73.3%)显著高于有淋巴结转移者(45.5%)(P<0.05).大肠癌中nm23-H1表达与肿瘤分化程度呈正相关(r =0.192,P<0.05),与肿瘤浸润深度呈负相关(r =-0.263,P<0.05).nm23-H1表达程度与患者年龄、性别、肿瘤大体分型、组织学类型、同期肿瘤大小均无关.结论:nm23-H1表达下调是大肠癌发生、发展中的重要分子生物学事件,可作为提示肿瘤分化、侵袭和淋巴结转移的一项指标. AIM: To investigate the expression of nm23-H1 and its correlation with the clinicopathological characteristics in human colorectal carcinoma, and to explore the role of nm23-H1 expression in the carcinogenesis, development and metastasis of colorectal carcinoma. METHODS: S-P immunohistochemical technique was used to detect the expression of nm23-H1 in 63 cases of colorectal cancer (including cancer and cancer-adjacent tissues), 16 cases of adenoma and 25 cases of normal colorectal tissues. RESULTS: The level of nm23-H1 expression was significantly lower in colorectal cancer tissues than that in cancer-adjacent, normal mucosal or adenoma tissues (58.7% vs 90.5%, 96.0%, 93.8%, P 〈 0.05), as well as in colorectal cancer patients with lymph node metastasis than thatin patients without metastasis (45.5% vs 73.3%, P 〈 0.05). The expression of nm23-H1 was positively correlated with the differentiation degree (r = 0.192, P 〈 0.05), but negatively correlated with the depth of invasion (r = -0.263, P 〈 0.05). Nm23-H1 expression had no relationship with the sex and age of patients', the morphologic type, histological type and sizes of the tumors. CONCLUSION: Down-regulation of nm23-H1 expression is an important molecular biological event in the carcinogenesis and progression of colorectal carcinoma, and can be used as an index for the judgment of tumor cell differentiation, invasion and lymph node metastasis.
