摘要 目的:了解RNA干扰沉默STAT3基因对结肠癌细胞侵袭的可能作用。方法:根据STAT3基因特点,设计合成STAT3小干扰RNA(small interfering RNA,siRNA),并转染人结肠癌细胞系HCT116后,采用RT-PCR检测STAT3mRNA,采用软琼脂集落培养试验检测癌细胞锚着不依赖性增殖,采用Boyden小室模型试验检测癌细胞的侵袭能力。结果:STAT3 siRNA可有效抑制结肠癌细胞集落生长和侵袭能力,且与浓度相关。结论:STAT3 siRNA可抑制结肠癌细胞的侵袭。 Objective: To study the effects of STAT3 small interfering RNA (siRNA) on invasion of colon cancer cell line HCT116. Methods: After colon cancer HCT116 cell were transfected by STAT3 siRNA, we studied the expression of STAT3 mRNA using RT-PCR, anchorage-independent growth using clon formation in soft agar, and invasion using hoyden chamber mode. Results: STAT3 siRNA could knock-out the level of STAT3 mRNA. STAT3 siRNA can inhibit anchorage-independent growth and invasion ability in a dose-dependent manner. Condusion: STAT3 siRNA can inhibit anchorage-independent growth and invasion ability of colon cancer cell.
