摘要
目的研究体外不同浓度的明胶类血浆代用品(Haemaccel)对正常人体免疫系统白介素-2,白介素10(IL-2,IL-10)及干扰素-γ(IFN-γ)的影响。方法正常志愿者6例,严格无菌条件下抽取静脉血各15ml,肝素抗凝,计数后将细胞平均分成5组:1640培养液阴性对照组、10%血浆对照组、10%血浆代用品试验组、20%血浆代用品试验组、40%血浆代用品试验组,然后再将各组分成阴性对照组、加脂多糖(LPS)组、加植物血凝素(PHA)组,使单个核细胞在不同的培养液中培养48h后,离心提取上清液,以酶联免疫吸附法(ELISA法)检测各上清液中细胞因子IL-2,IL-10和IFN-γ的浓度。结果三个浓度的血浆代用品组与1640培养液及血浆对照组在三种环境(阴性对照、加LPS、加PHA)中比较差异无统计学意义(P>0.05);在IL-2和IFN-γ,阴性对照组与加LPS组差异无统计学意义(P>0.05),而二者均与加PHA组差异有统计学意义(P<0.05);在IL-10,阴性对照组、加LPS组及加PHA组之间差异均有统计学意义(P<0.05)。结论明胶类血浆代用品(Haemaccel)在体外试验中不抑制正常人体免疫系统机能,其应用于肿瘤患者围术期以减少异体输血、保护其免疫状态的是安全可行的。
Objective To observe the influence of colloid plasma substitutes (Haemaccel) at different concentrations of IL-2, 10 and IFN-γ in normal population. Methods 6 healthy volunteers supplied venous blood 15 ml, the blood was anticoagulated by heparin. In super clean bench, single nuclear cells were separated from the blood. After being counted, the blood samples were divided into five groups; 1640 culture medium negative control group, 10% plasma group, 10% Haemaccel group, 20% Haemaccel group and 40% Haemaccel group. Then each group was divided into negative control group, adding LPS group, adding PHA group. The cells were cultured in each culture medium for 48 hours. After centrifuge, the superior clear fluid was extracted from the culture medium. Enzyme - linked immunosorbent assay (ELISA) was used to detect the levels of IL-2, IL-10 and IFN-γ in superior clear fluid. Results Compared trial groups with control groups in the three conditions (negative control, adding LPS, adding PHA), it showed that although the levels of cytokines in each group were different, there weren't significant difference among the groups (P〉0.05). In IL-2 and IFN-γ, there wasn't significant difference between negative control group and adding LPS group (P〉0.05), but there had significant difference between the two groups and adding PHA group (P〈0.05). In IL-10, there had significant difference among negative control group, adding LPS group and adding PHA group (P〈0.05). Conclusion Haemaccel doesn't restrain immune system of healthy persons in vitro. Haemaccel is a reasonable plasma substitute for cancer patients with compromised immune function compared with that in blood infusion perioperation.
