摘要
目的:初步探讨p53基因家族新成员——截断型 p63(ΔNp63)在小鼠生殖中的作用。方法:收集动情周期各期和早孕2 d、3 d、4 d、5 d小鼠子宫内膜,共8组,每组10只小鼠。运用RT-PCR技术检测ΔNp63 mRNA的表达规律;免疫组织化学SP法检测小鼠子宫内膜中ΔNp63蛋白的表达情况。结果:1)RT-PCR:动情前期ΔNp63 mRNA明显高表达;动情间期和动情期表达减弱;动情后期表达极低;早孕期间小鼠子宫内膜ΔNp63 mRNA表达较未孕组高,并逐渐增强,孕5 d达高峰。2)免疫组织化学:ΔNp63蛋白在间质细胞胞浆表达,表达规律基本上与mRNA水平的表达规律相似。结论:ΔNp63在动情前期高表达,可能与子宫内膜间质细胞增殖、生长有关;在早孕期间高表达,说明它很可能进一步促进间质细胞分裂,顺利转化成蜕膜细胞,进而有利于提高窗口期子宫的容受性。
Objective: To investigate the roles of △ Np63 in the female mouse procreation process. Methods: Endometria of mice in the early phase of pregnancy and estrus cycle were collected and divided into 8 study groups, and every group included 10 mice. The expression of △ Np63 mRNA and protein in the endometria of mice were detected by RT-PCR and immunohistochemical technique respectively. Results: High level expression of △ Np63 mRNA was observed in the endometria at proestrus. Low level expression at diestrus and estrus, and the less expression at metestrus; △ Np63 mRNA was expressed strongly in the endometria in the early phase of pregnancy and markedly different from the nonpregnant groups. Moreover, the highest expression was detected at d 5. Expression of △ Np63 protein was primarily limited to the subluminal stromal cytoplasm in the endometrium of mouse. Expressional mode of A Np63 protein was similar with that of the mRNA. Conclusion: △ Np63 expression clearly increases in the endometrium ofproestrus, which may enhance the proliferation and cleavage of the stromal cells. It is expressed strongly in the endometrium of early pregnancy that may promote the capability of uterus by accelerating further cleavage development and decidualization of the stromal cells.
