硫酸化魔芋葡甘低聚糖的抗乙肝病毒作用 被引量:3

摘要 目的体外观察硫酸化魔芋葡甘低聚糖(HOGS)抗乙肝病毒(HBV)的作用。方法采用四甲基偶氮唑盐(MTT)法和ELISA法,研究HOGS对HepG2—2.2.15细胞细胞毒性作用及其分泌HBsAg、HBeAg的影响。采用荧光探针定量PCR检测HOGS对HepG2-2.2.15细胞HBV—DNA复制的影响。结果HOGS在0.125~1mg/mL浓度范围对HepG2-2.2.15细胞没有明显的细胞毒性作用,细胞的半数中毒浓度(TC50)为1.4753mg/mL;并且可明显抑制HepG2—2.2.15细胞HBsAg的分泌,其半数有效浓度(IC50)为0.0268mg/mL,治疗指数达5.52;对HBV-DNA也有一定的抑制作用。结论HOGS具有一定的体外抗HBV作用。 Purpose To evaluate the inhibition effects of hydroxylpropyl oligo-konjac glucomannannuroate sulfate (HOGS) against HBV in vitro. Methods MTT assay was employed to detect the in vitro cytotoxicities of HOGS. HepG2-2.2.15 cell was used as the target cellular line. The effects of HOGS on the secretion of HBsAg and HBeAg were assayed by ELISA method. FQ-PCR technique was used to study the inhibitory effects of HOGS on the content of HBV-DNA in HepG2-2.2.15 ceils. Results The results showed that HOGS had no obvious cytotoxicities in the concentrations from 0. 125 mg/mL to 1 mg/mL and its TC50 was 1. 475 3 mg/mL. The results also showed that HOGS exhibiteda a strong inhibitory activity against the secretion of HBsAg, its IC50 was 0.0267 5 mg/mL and its therapeutic index(TI) was 5.52. HBV-DNA was inhibited effectively as well. Conclusion All these results indicated that HOGS had an inhibitory effect on HBV in vitro.
