摘要
目的观察养心颗粒(YXG)对急性,心肌梗死犬血管内皮细胞及心肌损伤的保护作用。方法采用结扎冠脉前降支的方法,复制急性,心肌梗死模型。分别测定冠脉结扎前以及药后不同时间点血浆超氧化物歧化酶(SOD)、丙二醛(MDA)、内皮素(ET)、一氧化氮(NO)、P-选择素及肌酸磷酸激酶同工酶(CK-MB)的变化。采用N-BT染色法测定心肌梗死面积。结果YXG治疗组与模型组比较,血浆CK-MB、MDA、ET、P-选择素降低(P〈0.05—0.01),而血浆SOD和NO升高(P〈0.05-0.01),梗死面积明显缩小(P〈0.01)。结论YXG对急性,心肌梗死犬心肌及血管内皮细胞的损伤具有保护作用,其作用机制可能与抗氧化、改善血管内皮细胞的分泌功能及抑制血小板活化有关。
Purpose To study the protective effect of Yangxin granules on vascular endothelial cells and myocardial injury in mongreldogs with myocardial infarction. Methods A total of 36 mongrel dogs were randomly divided into six groups,namely sham-operated group,myocardial infarction(MI) group,diltiazem group, and Yangxin granules 5,15,30 mg/kg groups.The experimental animals were handled by ligating the left coronary artery at the anterior descending branch, and a blood sample was withdrawn to measure the level of plasma ereatine kinase isoenzyme (CK-MB), the activity of superoxide dismutase (SOD), the content of malondialdehyde (MDA) and nitric oxide(NO) and the concentration of plasma endothelin(ET) and P-selectin. Infarction size was also determined. Results Infarction size was significantly less in Yangxin granules 15,30 mg/kg groups and diltiazem group compared with MI group( P 〈 0.01 ). Plasma CK-MB activity( P 〈 0.05 - 0.01 ) was significantly reduced in Yangxin granules 15,30 mg/kg groups vs MI group.Plasma MDA,a product of lipid peroxidation,was significantly decreased( P 〈 0.05) in Yangxin granules 15,30 mg/kg groups vs MI group,cansistent with the increased plasma SOD activity( P 〈 0.01 ). The level of plasma NO( P 〈 0.01) was comparably increased in Yangxin granules treated groups vs MI grou'p. Plasma ET and P-selectin concentrations were also decreased markedly( P 〈 0.05-0.01 ), compared with MI group. Conclusion Yangxin granules could effectively protect vascular endothelial cells and myocardial injury in mongrel dogs with myocardial infarction.
