The Effect of Adrenomedullin on Apoptosis in Activated Hepatic Stellate Cells of Rat.

摘要 目的探讨肾上腺髓质素(ADM)对活化肝星状细胞(HSC)凋亡的作用。方法大鼠肝星状细胞系(HSC-T6),以10%小牛血清DMEM培养基体外培养。采用不同浓度ADM单独以及与TNF-α一起对培养的HSC-T6进行干预,通过流式细胞术检测HSC-T6的凋亡率。结果ADM10-8mol/L组、10-7mol/L组、10-6mol/L组HSC-T6凋亡率分别为(8.81±0.27)%、(7.99±0.36)%、(5.61±0.33)%;空白对照组HSC-T6凋亡率为(10.82±0.38)%;ADM不同浓度组凋亡率均显著低于空白对照组(P<0.05)。TNF-α+0.9%NS组HSC-T6凋亡率为(24.53±2.81)%,ADM低、中、高浓度+TNF-α组凋亡率分别为(21.80±3.35)%,(14.43±2.13)%,(12.96±1.61)%;各浓度ADM+TNF-α组凋亡率与TNF-α组相比差异有统计学意义(P<0.05)。结论肾上腺髓质素可呈剂量依赖性直接或间接通过干预TNF-α的作用抑制活化HSC的凋亡。 Objective To investigate the effect of adrenomedullin( ADM )on the apoptosis in the activated hepatic stellate cells of rat in vitro. Methods Hepatic stellate cells of rat ( HSC-T6 ) was cultured in DMEM medium with 10 % calf serum in vitro. The apopto- sis rat of HSC-T6 was observed by flow cytometry after administration of different concentrations ADM with or not with TNF-α. Results The apoptosis rate of HSC-T6 were(8.81 ± 0.27) %, (7.99 ± 0.36) % and(5.61 ± 0.33) % in ADM groups respectively,which are all lower than that in control group( 10.82 ± 0.38) % ( P 〈 0.05) .The apoptosis rate of HSC-T6 were(21.80 ± 3.35) %, ( 14.43 ± 2.13) % and (12.96± 1.61)% in TNF-α and different concentrations ADM groups respectively, whiche are all lower than that in TNF-α and 0.9 % NS group (24.53 ± 2.81 ) % ( P 〈 0.05 ). Conclusion ADM can directly or indirectly decrease the apoptosis rate of the activated HSC of rat,and the apoptsis rate was correlated with dosage of ADM in negatively.
