摘要
目的:比较不同大小的力对腺样囊性癌高低转移细胞系增殖的影响。方法:通过体外培养腺样囊性癌高低转移细胞系ACC-2,ACC-M,建立双轴力学刺激细胞模型,用不同大小力刺激,用流式细胞仪检测ACC-2,ACC-M增殖变化。结果:不同大小的力及作用时间对ACC细胞增殖活性的影响具有统计学意义,并且这种影响还具有随时间延续而变化的特性。结论:力对细胞增殖有影响并与力大小和作用时间相关。
Objective: To investigate the effects of cyclic biaxial mechanical stimulation on the proliferation of adenoid cystic carcinoma(ACC) cell lines ACC-M and ACC-2. Methods:ACC-2 and ACC-M cells were harvested. Mechanical stimulations were applied on the cells in vitro using a cyclic biaxial mechanical stimulation device, and the stimulations were respectively applied to the cells for periods of 1,3 and 6 hours to each group respectively, once a day for 2 days. The amplitude of mechanical stimulations applied to the cells were 4 000 and 1 000 Ixstrain respectively, at a frequency of one hertz ( 1 Hz). Unstrained cells were used as the control The proliferation of the ceils was studied with flow cytometry. Results: The proliferation index(PI) of ACC-M cells was increased by 1 h-stimulation,decreased by 3 h-stimulation at 1 000 or 4 000 μstrain;decreased at 1 000 μstrain but increased at 4 000 μstrain by 6 h-stimulation. PI of ACC-2 cells was increased by 1 h-stimulation at 1 000 or 4 000 μstrain, decreased by 6 h-stimulation at 1 000μstrain and 3 h-stimulation at 4 000 ixstrain, but increased by 6 h-stimulation at 4 000 μstrain. Conclusious:The data prove that in vitro mechanical stimulations can influence the proliferation of ACC-2 and ACC-M cells.
