氧化低密度脂蛋白对培养兔脂肪细胞凝血和纤溶因子表达的影响 被引量:2

摘要 目的探讨氧化低密度脂蛋白(ox-LDL)对培养的兔脂肪细胞表达组织因子(TF)和纤溶酶原激活物抑制剂1(PAI-1)的影响。方法取正常兔脂肪组织分离培养脂肪细胞,以不同浓度(分别为0、5、25、50、75μg/ml)ox-LDL孵育兔脂肪细胞24h后收集细胞。RT-PCR测定脂肪细胞TF和PAI-1 mRNA表达。用ELISA方法测定细胞裂解液中TF蛋白和培养上清中PAI-1浓度。结果ox-LDL可增加兔脂肪细胞TF和PAI-1的表达和蛋白产生。在ox-LDL50μg/ml培养时兔脂肪细胞TF和PAI-1 mRNA分别为0.844±0.015和0.672±0.022;ox-LDL75μg/ml培养时分别为0.939±0.015和0.846±0.012,与对照比较(分别为0.763±0.013和0.529±0.013)明显增加(P<0.01)。结论ox-LDL能刺激脂肪细胞TF和PAI-1 mRNA表达,并使其蛋白生成增加,提示ox-LDL促进脂肪细胞TF和PAI-1分泌,继而影响血栓形成。 Objective To investigate the effects of oxidized LDL (ox-LDL) on tissue factor (TF) and plasminogen activator inhibitor-1 (PAI-1) mRNA expression in cuhured adipocytes. Methods Adipocytes from normal rabbits were incubated with increasing concentration of ox-LDL (0, 5, Z5,50, 75μg/ml)for Z4 hours. TF and PAI-1 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). TF and PAI-1 antigen were determined with ELISA. Results ox-LDL significant- ly enhanced the mRNA expression and protein production of TF and PAI-1. With the culture of ox- LDL 50μg/ml, the expression of TF and PAI-1 mRNA were 0. 844±0. 015 and 0. 672±0. 022, respectively; and were 0. 939±0. 015 and 0. 846±0.012 with the cuhure of ox-LDL 75big/m1, respectively, which were higher than those of the controls (0. 763±0. 013 and 0. 529±0. 013, respectively, P〈0. 01). Conclusion ox-LDL stimulates the expression of TF and PAIl and enhances the secretion of TF and PAI-1 in adipocytes, it may promote thrombosis formation.
