摘要
在葡萄糖氧化酶发酵过程中,配以外置式的膜过滤系统进行膜过滤发酵,可减缓发酵过程中产物及分解代谢物对酶合成的阻遏和抑制作用,使酶产率增加。实验结果表明:膜过滤发酵的产酶速率高达4.140×16.67μmol/s·h(4140.19U/h),比分批发酵高3倍以上,且能在较长时间内维持较高的酶产率。膜过滤发酵过程中pH、NH_2-N和葡萄糖酸的含量随补糖速率和稀释率D的大小发生变化,发酵液中葡萄糖含量控制在2.0%~2.5%,D控制在0.12h ̄(-1)为宜。
Glucose oxidase(GOD)fermentation in stirred tank bioreactor coupled with a membrane filtration system is evaluated in order to remove the catabolites inhbition and repression and to increase the enzyme productivity.Experimental results show that the maximum enzyme productivity is 4.14×16. 6μmol/s·h(4140.19U/ h),which is 3 times higher than that of the batch fermentation.The operation conditions for membrane filtration are also studied.The optimum glucose concentration in broth is controlled within 2.0% ~2.5%,The optimum dilution rate is 0.12h~(-1).
