Expression of nerve growth factor in painful traumatic neuroma and its significance

摘要 目的 观察神经生长因子(nerve growth factor,NGF)在创伤性神经瘤中的表达与分布。探讨其与创伤性神经瘤疼痛的关系和意义。方法 将51例创伤性神经瘤组织标本,分为创伤性神经瘤无疼痛症状组(无疼痛组,36例),创伤性神经瘤有疼痛症状组(疼痛组,15例);并将15例正常神经组织标本,设为对照组。采用免疫组化方法辅以计算机图像定量分析技术,对NGF在创伤性神经瘤中的表达进行观察和检测。结果 对照组未见NGF的阳性表达,其它2组皆见阳性表达。但有疼痛症状组NGF阳性表达明显强于无疼痛组,t检验显示两者差异有统计学意义(t=54.71,P〈0.01);Spearman等级相关分析表明,NGF的表达强度与疼痛分数呈正相关关系(r8=0.91,P=0.00)。其直线回归方程为:Y=7241,74X-57.43(P=0.00)。结论 NGF是导致创伤性神经疼痛的一个重要原因。 Objective To observe the expression and distribution of nerve growth factor (NGF) in traumatic neuroma and discuss its significance and relationship with neuroma pain. Methods Traumatic neuroma samples were collected from 51 cases, 36 of which had pain while 15 didn't have pain. Fifteen normal nerve samples served as control. Immunohistochemical staining was performed to detect expression of NGF in the neuroma and quantitate the expression levd by using computer imaging analysis system. Results There was no expression of NGF in samples of the control group. Expression of NGF was seen in neuromas of both the pain group and no pain group. The expression level was higher in the pain group than that of the no pain group, the difference being statistically significant ( t = 54.71, P 〈0.01 ). Correlation analysis indicated that the expression level of NGF was positively correlated with the scale of the pain assessed by means of a 10-point visual analogue scale (VAS) (r8 = 0.91, P = 0.00)and the linear regression equation was Y = 7241,74X - 57.43 ( P = 0.00 ). Conclusion NGF plays an important role in the cause of traumatic neuroma pain.
