摘要
目的研究粉防己碱(tetrandrine,Tet)对大鼠角膜新生血管(corneal neovasculari-zation,CNV)缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法建立大鼠角膜碱烧伤诱发CNV模型,采用裂隙灯照相、免疫组化和RT-PCR等方法,分别检测使用Tet前后HIF-1α和VEGF在SD大鼠CNV中的表达情况。结果在正常角膜,HIF-1α和VEGF没有表达或在上皮基底膜有微弱表达;在新生血管对照组,角膜全层HIF-1α和VEGF表达明显增强,其表达部位主要分布在新生血管形成区域和上皮全层;用药组角膜新生血管密度明显减低,HIF-1α和VEGF相应在角膜基质层新生血管形成区域表达减少。经SPSS软件分析,3组间具有统计学差异。结论Tet可以有效地抑制大鼠角膜碱烧伤诱发CNV的生长,降低角膜新生血管HIF-1α和VEGF的表达。
Objective To investigate the effect of tetrandrine (Tet) on the expression of hypoxia inducible factor-1α ( HIF-1α ) and vascular endothelial growth factor (VEGF) in corneal neovasction(CNV). Methods CNV model was made by alkaline burning. The expressions of HIF-1α and VEGF in CNV before and after using Tet were detected by slit lamp biomicroscopy, photography, immunohistochchemical staining and RT-PCR. Results The expression of HIF-1α and VEGF in normal rat cornea was negative or very weak;But there was strong expression in the region of CNV formation and all Myers of corneal epithelium mainly in control group of CNV. In therapy group of Tet,the expressions of HIF-1α and VEGF in corneal stroma were weak.There were were signifcant differences among three groups with SPSS software. Conclusion Tet can inhibit the growth of CNV induced by alkaline burning in rats effectively,and reduce the expression of HIF-1α and VEGF in CNV. [ Rec Adv Ophthalmol 2007 ;27 (2) : 102-105 ]
