摘要
本文报道用固体培养法培养坛紫菜叶状体营养细胞试验的结果。试验表明,坛紫菜叶状体细胞可以在半固态至固态琼脂培养基上良好地存活、分裂和再生,因此用固体培养法进行其细胞培养的有关研究是可行的。细胞在半固态培养基中的分裂再生情况与在液培中完全一致。在固体培养基表面培养情况亦基本相似,单离细胞分裂与再生形成的产物亦有正常苗、畸形苗、细胞团、丝状体等,但产生的细胞团较多,无精子囊器形成;再生苗可在培养基上持续缓慢地生长,丝状体可正常地快速生长并发育成熟;观察到部分细胞团经培养发展成愈伤组织,后者可分化长出叶状体;个别单离细胞经培养直接萌发成壳孢子囊枝。本研究还观察了Ca ̄(++)浓度对细胞再生与发育情况的影响,结果表明Ca ̄(++)浓度低的培养基上长出的叶状体多、丝状体少、并且生长差、发育慢。
篢his paper reports the result of the cultivation of vegetative cells from Por-phyra haitanensis(Rhodophyta)by using solid culture method.The result of experimentsshows that the vegetative cells can survive,divide and regenerate well on semisolid andsolid agar seawater medium. The division and regeneration of cells on semisolid mediumis much the same as that in liquid medium. The cells isolated divided and regenerated toform thalli(with or without holdfast),conchocelis all but more cell-groups and no sper-matangia on solid medium are similiar to that in liquid medium. On solid medium,thalliregenerated can slowly and continuously grow,and conchocelis can normally grow andmaturate. throughout culture,parts of cell-groups can develop into callus and furthurdifferentiate into thalli; few cells isolated can derectly sprout into conchosporangiabranch.The experiment that Ca++ concentration affects regeneration and growth of cellsisolated shows that vegetative cells grow into more thalli and less conchocelis on lowerCa++ concentration contained solid medium. Furthermore,the conchocelis grow badly andslowly.
