摘要
目的:建立用对二甲氨基苯甲醛比色法分析保健食品中胶原蛋白的检测方法。方法:样品在氯化亚锡的盐酸溶液中水解,释放出羟脯氨酸,经氯胺T氧化,羟脯氨酸氧化物与对二甲氨基苯甲醛反应生成红色化合物,在波长560nm处测吸光度。结果:标准曲线相关系数r=0.9997;变异系数〈5%;加标回收率在90.0%~93.1%之间;检出限为1.5μg/ml;羟脯氨酸浓度在2~23μg/ml范围内与吸光值呈线性关系。结论:本方法测定的线性范围广,相关性、重现性好,准确度高,具有较高的实用价值。
Objectlve:To establish a method of p - Dimethylaminobenzaldehyde colorimetry for determination collagen in health foods. Methods:The sample was hydrolyzed by SnCl2 - HCI solution, hydroxyproline was released and oxidized by chlora- mine - T. Hydroxyproline oxide with p - Dimethylaminobenzaldehyde could form red compounds, their absorbency was detected at the wavelength of 560 nm. Results :The correlation coefficient of calibration curve was 0. 9997, coefficient of variation less than 5%, the recovery was 90.0% -93. 1%, the limit was 1.5 μg/ml. Collagen showed a good linear relationship with absorbance when hydroxyproline concentration was between 1 -23 μg/ml. Conclusion:The p -Dimethylaminobenzaldehyde colorimetry is a good method with wide linear range, well correlation and repetition ,high accuration and practicalbility.
