摘要
应用间接免疫荧光法分析了人天然抗体与猪胰腺细胞的反应。18份胰岛素依赖型糖尿病患者的血清及20份正常人血清分别与杂种猪胰腺组织的冰冻切片孵化后,用免疫荧光标记的羊抗人 IgG、IgM 单克隆抗体染色。结果显示,所有患者的血清及正常人血清均能与一种或多种胰腺细胞反应,这些细胞包括胰岛细胞、血管内皮细胞、导管上皮细胞及单核巨噬细胞。认为人体内存在着抗猪胰岛细胞的天然抗体,体液免疫机制在异种猪胰岛移植的排斥反应中可能起了重要作用;临床移植前有必要进行配合试验和移植物预处理。
The reactivity of XNAs in the serum samples from 18 insulin-dependent diabetes mellitus(IDDM)patients and 20 healthy persons to the pancreatic tissues from hybrid pigs was studied by indirect immuno-fluorescence staining techniques.The frozen sections of the pancreas were incubated with the sera from diabetic patient or human controls,and then incubated with FITC-conjugated goat antihuman IgG or IgM monoclonal antibodies.The results showed that all IDDM and human control sera could react to one or the other kinds of the pancreatic cells which include:islet cells,vascular endothelial cells,ductal epithelial cells and macrophages.It is sug- gest that 1.IDDM patients and healthy persons contain NXAs against porcine pancreatic islets. The humoral immunity might play a major role in rejection in porcine islets xenotransplantation. 2.Cross-matching test and pretransplant graft treatment are needed pretransplantly.
