摘要
我院于1992年研制成长征-1多器官保存液(简称 CZ-1液)。同时对心肌组织0、18、24、36、48小时冷冻保存后的形态学作了观察,并与 UW 液作了比较。结果表明:(1)从光镜观察来看,24小时后心肌细胞浊肿,部分横纹不清。肌纤维紊乱,有断裂。肌纤维间隙增宽。48小时后心肌细胞散存性坏死。其 CZ-1液组与 UW 液组无明显区别。(2)从电镜观察来看,18小时后心肌细胞受到轻-中度损伤。线粒体肿胀,部分嵴膜消失,出现"气球样"。24小时后心肌细胞受到严重的损伤。而 UW 液保存组其心帆组织保存18小时以后电镜下超微结构的损伤似乎重于 CZ-1液。我们认为,自制的 CZ-1液能较为有效地保存兔心脏在18小时以内。
Rabbit heart tissues cold stored with CZ-1 solution were histologically investigated and were compared with those stored with UW solution.Under light microscope,24h after cold storage,the cardiac muscle cells were found to have cloudy swelling with muscle fiber disarranged or broken.The spacing between fibers were widened.48h after storage,cardiac muscle cells showed scattered necrosis.No significant histological difference was found between CZ-1 and UW groups.Under electron microscope,18h after storage,the cardiac muscle cells revealed mild to moderate injury.The mitochondria were swelling with the ballooning degeneration or crest disap- pearing.24h later,the myofibrils were broken and lysed;the mitochondria became swelling and ruptured with liquefactive degeneration and pyknosis.The morphological changes in CZ-1 solution group were slighter to UW solution group within 18h of cold storage.This showed that within 18h of cold storage,CZ-1 solution is effective in the preservation of rabbit heart.
