摘要
目的研究僵蚕提取液的抗凝活性成分的稳定性和提取工艺。方法以凝血酶时间(TT)为指标,考察僵蚕提取液中抗凝物质对酸、碱、热及反复融冻的稳定性;以紫外分光光度法测其蛋白含量为指标,用L9(34)正交试验考察僵蚕的提取工艺。结果僵蚕水提液的抗凝活性成分对酸、碱、热及反复融冻均具有较高的稳定性;僵蚕水提液的凝血酶时间与蛋白质含量成正相关(P<0.01);以蛋白质含量作为指标,僵蚕用8倍量水润湿15 min,提取2次,每次30 min,即为水煎提取优化工艺。结论为僵蚕抗凝活性成分的水煎提取和后续分离的样品处理提供了实验依据。
Objective To investigate the stability of anti-coagulate active components (AUAU) in extracted liquor from Bombys Batryticatus (ELBB) and its extract technique. Methods The stability of ACAC under hot, acid, alkali or a series of frozen-thaw were observed based on the determination of thrombin time (TT); the extract technique of ELBB were estimated with the L9 (3^4) orthogonal experiment based on the contents of total proteins by ultravelet spectrophotometry. Results ACAC of ELBB was provided with a good stability even though under hot, acid, alkali or a series of frozen-thaw; TT of extracted liquor from ELBB was correlated with the contents of total proteins (P〈0.01); the optimized extract technique was that the Bombys Batryticatus was wetted 15 min. in 8 times of water, extracted 2 times in 30 minutes according to the index of protein content. Condution The study may provide a experimental basis for extraction and later separation of ACAC from Bombys Batryticatus.
