摘要 目的探讨三氧化二砷(As2O3)对人宫颈癌Hela细胞系的放射增敏作用。方法MTT法确定As2O3对Hela细胞的半数致死浓度(LD50),采用集落形成法观察20%该浓度的As2O3对Hela细胞的放射增敏作用。结果(1)细胞生长抑制随As2O3浓度的增加而增强;(2)As2O3+照射组的细胞存活率低于单纯照射组,D0值小于单纯照射组(1.58Gy、2.11Gy),Dq值也小于单纯照射组(0.27Gy、0.64Gy),存活曲线肩区(Dq)变小,放射增敏比(SER)1.34。结论As2O3对宫颈癌细胞有明显的放射增敏作用,其作用机理有待进一步研究。 Objective To investigate the irradiation enhancement effect of arsenic trioxide ( As2O3 ) on human cervical squamous carcinoma cells. Methods The concentration LD5o of As2O3 was determined by MTT, the irradiation enhancement effect was determined by cloning assay with the concentration 20% of LD50. Results ( 1 ) Inhibitition of cell proliferation seemed more dependent on the increase of As2O3 concentration. (2) Cell survival rate was lower in the combination of As2O3 and radiation than that of As2O3 alone, Dq、D0 value was decreased in the combined As2O3 and radiation than that of radiation only(Dq:0.27 Gy vs 0.64 Gy', DO 1.58 Gy vs 2.11 Gy), with radiation enhancement ratio of 1.34. Conclusion As2O3 is able to enhance radiation effect obviously on human cervical squamous carcinoma cells.
