胰岛素样生长因子-1在病毒性心肌炎小鼠中的变化及对心肌的保护作用 被引量:6

Changes of Insulin-Like Growth Factor-1 in Mice with Viral Myocarditis and Their Protective Effects on Myocardium

摘要 目的探讨病毒性心肌炎(VMC)小鼠血清胰岛素样生长因子-1(IGF-1)水平的变化及外源性IGF-1对凋亡相关蛋白Bcl-2和Bax表达的影响。方法Balb/c雄性小鼠随机分为3组:正常对照组、柯萨奇B3病毒(CVB3)感染组(感染组)和病毒感染加IGF-1治疗组。分别在实验d7、d14处死动物,留取血清、心肌标本。放射免疫法检测血清IGF-1水平,免疫组织化学法检测心肌组织Bcl-2和Bax蛋白表达。结果感染组血清IGF-1水平较对照组明显降低,Bcl-2表达减少、Bax表达增加(P<0.01);治疗组较感染组Bcl-2表达增加、Bax表达减少(P<0.01)。结论IGF-1对VM小鼠的心肌细胞有保护作用。 Objective To determine the significance of change of insulin - like growth factor - 1 ( IGF - 1 ) and study the effect of IGF - 1 on expression of Bcl - 2 and Bax, Methods Male Balb/e mice were randomly divided into control group, eoxsaekie B3 ( CVB3 ) infection group (infection group) ,CVB3 infection with IGF- 1 treated group. Animals were killed at d7, d14, The immunohistechemieal studies and medical imagine analysis system were performed to evaluate the expression of Bcl -2 and Bax in myocardial tissue. Serum IGF - 1 concentrations were assayed by radioimmunoassay(RIA), Results In infection group, the concentrations of IGF - 1 were more lower than that of control group and the expression of Bcl - 2 reduced and Bax increased( P 〈 0.01 ). As compared to infection group, the expression of Bcl - 2 increased and Bax reduced in treated group ( P 〈 0, 01 ). Conclusion IGF - 1 may offer protection to cardiomyecyte of viral myecarditis in mice.
