摘要 目的研究胰岛素样生长因子结合蛋白5(IGFBP-5)在高体积分数氧致新生大鼠慢性肺疾病(CLD)中的表达及作用机制。方法将足月新生大鼠96只随机分为高体积分数氧组(高氧组)和空气组,分别于实验1、3、7、10、14、21d应用免疫组织化学和RT—PCR技术检测IGFBP-5的动态表达及阳性表达部位。结果CLD时IGFBP-5呈动态变化。高氧组和空气组比较,在实验3~10d IGFBP-5表达明显增强,14051121d表达明显降低。阳性部位在支气管上皮细胞、肺泡上皮细胞、成纤维细胞、血管周围和间隔的间质细胞。结论IGFBP-5参与CLD发生,作为胰岛素样生长因子-I的抑制因子发挥作用。 Objective To investigate the expression of insulin - like growth factor - binding protein - 5 ( IGFBP - 5 ) in full - term rats with hypemxia - induced chronic lung diseases(CLD) and its mechanism. Methods Ninety - six full - term rats were randomly exposed to hypemxia ( hypemxia group) and room air( room air group). CLD was induced by hypemxia exposure. RT - PCR and immunohistochemieal methods were used to detect the expression of IGFBP - 5 at 1,3,7,10 , 14 and 21 days after exposure. Results The expression of IGFBP - 5 was dynamic, the expression of IGFBP -5 increased significantly in hypemxia group compared with room air group at 3 d to 10 d after hypemxia exposure ( P 〈0.05) ,otherwise the expression of IGFBP -5 decreased significantly in hypemxia group compared with room air group at 14 d and 21 d after hypemxia exposure ( P 〈 0. 05 ). IGFBP - 5 was immunolocated mainly in bronchial epithelial cells, alveolar epithelial cells, fibmblast and interstitial cells. Conclusions IGFBP - 5 is involved in CLD. IGFBP - 5 acts as a inhibitory factor to insulin - like growth factors - I in the development of CLD.
