摘要
目的:探讨高迁移率族蛋白B1(high mobility group box chromosomal protein 1,HMGB-1)在小鼠巨细胞病毒(murine cytomegalovirus,MCMV)感染导致听力损失发病机制中的作用。方法:采用新生小鼠颅内注射病毒法建立MCMV感染动物模型,测定2周龄小鼠的听性脑干反应(auditory brainstem response,ABR),采用RT-PCR检测耳蜗组织HMGB-1mRNA表达,均与注射生理盐水的对照组比较。结果:ABR,病毒组(28.42±3.03)dB,对照组(15.66±0.38)dB(t=3.042,P<0.01)。HMGB-1mRNA,病毒组2.372±0.193,对照组0.754±0.322(t=7.537,P<0.01)。结论:HMGB-1可能通过维持并加重内耳的迟发性炎症,成为巨细胞病毒感染导致听力损失发生发展的病理基础。
Objective To probe into the role of high mobility group box chromosomal protein 1 (HMGB-1)in the pathogenesis of hearing loss induced by infection of murine cytomegalovirus (MCMV). Methods MCMV-infected murine model was established by intracranial injection of newborn mice with viral suspension and the same volume of 0.9% sodium chloride, of which the latter was taken as the control group. The auditory brainstem response(ABR)and the mRNA level of HMGB-1 in the cochlea were assayed when the mice were 14 days old. Results The ABR thresholds was higher significantly than that in the control group [ (28.42 ±3.03) dB vs (15.66± 0.38) dB, t=3.042, P〈0.01 ]. The expression of HMGB-1 mRNA was also higher than that in the control group(2.372±0.193 vs 0.754± 0.322, t=7.537, P 〈 0.01). Conclusion HMGB-1 may be the important pathological basis of hearing loss induced by MCMV infection through sustaining and aggravating delayed inflammation in inner ear.
